The outer membrane of bacteria is made up of a complex lipid/sugar structure known as lipopolysaccharide (LPS) or endotoxin. This structure activates the Toll-like receptor 4 (TLR-4)-MD2-CD14 pathway, triggering an immune response in mammals.
While various methods have been developed to reduce endotoxins in many products, there is no universal approach that works for all products. This can be attributed to the diversity of endotoxin molecules, as only a portion of the population is removed by a reagent, or because the product and endotoxin share similar physical characteristics and cannot be separated from each other.
Endotoxin removal often results in increased loss of product, as well as the investment of time and reagents. The use of a genetically stable organism that reduces endotoxin contamination or facilitates easier removal is crucial as the production of gene therapies and mRNA vaccines continues to rise in the pharmaceutical industry.
Advances in Endotoxin Reduction with Clean Genome E. coli
Scarab Genomics has developed low endotoxin E. coli strains by eliminating the enzymes responsible for specific fatty acid chains in lipid A. These strains produce a plasmid that does not activate Toll-like receptor 4 in a mammalian cell culture model. Additionally, other strains have deletions in components of both the core polysaccharide and disaccharide diphosphates, resulting in strains that produce plasmid with very low endotoxin levels, as confirmed by the Limulus amebocyte lysate (LAL) assay.
