Clean Genome® E. coli strains are the bacterial hosts of choice for a wide spectrum of protein and nucleic acid production applications ranging from routine cloning and recombinant protein expression experiments to large scale production of nucleic acid and protein biopharmaceuticals.
Completely free of IS elements and cryptic prophages, plus deleted for many nonessential genes our range of Clean Genome® E. coli strains provide enhanced genetic stability and improved metabolic efficiency, to maximize protein and nucleic production efficiency. All strains are derived from the revolutionary MDS42 reduced genome strain described in Science 312:1044-6, and are further deleted to create the MDS69 series. Use of Scarab’s patented Clean Genome® strains requires a Limited License Agreement.
Ready for transformation, we offer six strains in the form of electrocompetent and chemically competent cells.
Clean Genome®
is the most robust bacterial fermentation host for general/routine recombinant protein expression and nucleic acid production
Clean Genome® ΔrecA
is an enhanced strain that is recombination deficient, suitable for production of nucleic acids with structures that may be unstable in recA+ E. coli, such as direct repeats, inverted repeats, stem-loops, etc.
Clean Genome® LowMut
further reduces the already low mutation rate of Clean Genome® strains. It has been engineered to reduce error-prone repair, thereby reducing the mutation rate to almost zero, even under the most stressful conditions.
Clean Genome® LowMut ΔrecA
combines the benefits of the ΔrecA and LowMut strains for maximum stability of otherwise troublesome nucleic acid structures, with the lowest possible mutant frequency.
Clean Genome® LowMut ΔrecA Blue
is Clean Genome® ΔrecA with the lacZ M15 genomic deletion to allow blue/white screening of inserts in plasmids using the α-complementing fragment of β-galactosidase.
Clean Genome® Low Mut T7
provides a T7 promoter in the genome for clients wishing to use this expression option rather than the synthetic T5 promoter in the recommended pSX2 vector.
Additional Strains
Should you have specific needs not addressed by any of the above strains, we have an extensive library of Clean Genome® E. coli strains with various additional genetic modifications, please contact support for more information.
Note: for optimal production efficiency, we strongly recommend the use of Korz minimal medium (see protocol for components) and strongly recommend against using M9 medium.
Custom libraries
